2 - 3 inches would be plenty, and laying down builders plastic (the thick
black stuff used in building foundations) first would be the best option.
You will never stop all weeds with just the gravel, neither will using the
plastic, but it helps.

I used weed mat under my gravel - seems to work a treat.

The previous owner had plastic in some areas of the backyard - the ants just
loved creating nests under there, not to mention the spiders!
